The adenovirus (AdV) icosahedral capsid encloses a nucleoprotein core formed by the dsDNA genome bound to numerous copies of virus-encoded, positively charged proteins. For an efficient delivery its genome, AdV must undergo cascade dismantling events from plasma membrane nuclear pore. Throughout this uncoating process, virion moves across potentially disruptive environments whose influence in particle stability is poorly understood. In work we analyze effect acidic conditions on particles exploring their mechanical properties, accessibility and disruption. Our results show that under short term acidification becomes softer less accessible intercalating dye, even presence openings. AFM tip penetrates deeper virions at neutral pH, properties genome-less are not altered upon acidification. Altogether, these indicate main compaction nucleoproteic core, revealing previously unknown role for chemical cues uncoating.
